General description
The mS2i6A is an evolutionally conserved modification found in the tRNA. The hypermodified mS2i6A at position 37 occurs frequently in tRNAs that read codons starting with uridine. The mS2 group of mS2i6A is critical for the accurate decoding of Tyr and Phe codons. It has been shown that a deficiency of mS2i6A in the cognate poly(U) reader tRNA(Phe) leads to increased misreading when the near-cognate competitor tRNA4(Leu) is wild-type. It can lead to higher rejection rate of tRNA4(Leu) by more aggressive proofreading on the wild-type ribosome. It is believed that Cdk5rap1 specifically catalyzes the mS2 modification of mt-tRNAs and contributes to mitochondrial function in vivo. This modification regulates efficient mitochondrial translation and energy metabolism in mammals. Cdk5rap1 knockout mice that are deficient in the 2-methylthio modification in mt-tRNAs bearing Ser(UCN), Phe, Tyr and Trp, impaired mitochondrial protein synthesis is observed, which causes respiratory defects of mitochondria and myopathy in vivo. Specificity
Clone CL-6 recognizes wild-type total RNA, but not Cdk5rap1 KO total RNA.
Immunogen
A synthesized mS2i6A nucleotide conjugated to BSA.
Application
Anti-mS2i6A, clone CL-6, Cat. No. MABS1280, is a mouse monoclonal antibody that detects mS2i6A and has been tested for use in Dot Blot and ELISA.
