Allgemeine Beschreibung

Protein polybromo-1 (UniProt: Q86U86; also known as hPB1, BRG1-associated factor 180, BAF180, Polybromo-1D) is encoded by the PBRM1 (also known as BAF180, PB1) gene (Gene ID: 55193) in human. BAF180 is a widely expressed, bromodomain containing nuclear protein that is an essential component of the SWI/SNF-B (PBAF) chromatin remodeling complex. It is involved in transcriptional activation and repression of select genes by chromatin remodeling. It contains six bromodomains, two bromo-adjacent homology (BAH) domains (aa 956-1074 and 1156-1272), and one HMG-box (aa 1379-1447). The bromodomains recognize and bind to acetylated lysines and BAH domains mediate protein-protein interaction. Its HMG-box domain binds to DNA. BAF180 acts as a negative regulator of cell proliferation. It plays a role in DNA repair mechanisms and acts as a tumor suppressor. It is also involved in silencing transcription near double-stranded DNA breaks (DSB). The BAH domains of BAF180 are shown to be essential for ubiquitination of PCNA. BAF180 depleted cells can not produce proper levels of ubiquitinated PCNA that leads to the inability to replicate past DNA lesions. Mutations in PBRM1 gene have been linked to high incidence of renal cell carcinoma. Nine isoforms of BAF180 have been described that are produced by alternative splicing. Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Protein polybromo-1 (BAF180) in Jurkat cell lysate.
