Human lung carcinoma, adrenal gland metastasis

Cell Line Description

The NCI-H510A cell line was derived by D. Carney, A.F. Gazdar amd associates in 1982 from an adrenal metastasis in an adult, 56-year-old, male Caucasian patient. The cells produce easily detectable p53 mRNA at levels comparable to those in normal lung tissue. The line does not exhibit any gross structural DNA abnormalities. The cells express elevated levels of four biochemical markers of small cell lung carcinoma (SCLC) i.e. neuron specific enolase, the brain isoenzyme creatine kinase, L-DOPA decarboxylase and bombesin-like immunoreactivity. This cell line is tumourigenic in nude mice (the cells form transplantable tumours with typical small cell carcinoma histology).

Culture Medium

RPMI 1640 + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:8 i.e. seeding at 2-4x10,000 cells/cm2; 5% CO2; 37°C. This cell line grows as a mixture of cells in suspension and adherent cells. Subcultures can be prepared by scraping the adherent cells into the medium,
