General description

Target of EGR1 protein 1 (UniProt Q96GM8) is encoded by the TOE1 gene (Gene ID 114034) in human. Originally characterized and thereby named as a downstream target of the immediate early gene Egr1, TOE1 is a nuclear protein localized primarily in nucleoli and Cajal bodies. TOE1 overexpression causes an upregulation of the cyclin-dependent kinase inhibitor p21 and cell cycle arrest at G2/M. TOE1 also interacts wtih p53 C-terminal tetramerization domain and enhances p53 transcriptional activity. In addtion, TOE1 exhibits inhibitory activity against HIV-1 replication and evidence indicates TOE1 interaction with the viral specific transactivator response element as part of the inhibitory mechanism. TOE1 is upregulated and secreted by activated CD8(+) T lymphocytes, and TOE1 can be proeolytically cleaved by granzyme B at D328 and D363, generating a 35-amino acid (a.a. 329-363) fragment containing the nuclear localization signal/NLS (a.a.335-347). The 35-a.a. fragment retains both the antiviral potency and cell-penetrating capability. Human TOE1 is produced as a 510-a.a. protein, containing a C3H1-type Zinc finger (a.a. 294-322) and anuclear localization signal/NLS (a.a.335-347). The initiation methionine is removed posttranslationally.

Specificity

This polyclonal antibody was raised against the C-terminal end sequence present in both spliced isoforms of human TOE1 reported by UniProt (Q96GM8).

Application

Immunoprecipitation Analysis: 1 µL from a representative lot immunoprecipitated TOE1 from 20 µg of HEK293T cell lysate (Courtesy of Dr. Ian de Belle).

Western Blotting Analysis: A representative lot detected FLAG-tagged wild-type or mutant TOE1 with D328A, D363A/D373A, or D387A point mutions following their immunoprecipitation by an anti-FLAG antibody (Sperandio, S., et al. (2015). Proc. Natl. Acad. Sci. U. S. A. 112(26):E3392-E3401).
