General description

Mis18-binding protein 1 (UniProt Q6P0N0; also known as hsKNL-2, Kinetochore null 2 homolog, Kinetochore-associated protein KNL-2 homolog, P243, Putative protein p243 which interacts with transcription factor Sp1) is encoded by the MIS18BP1 (also known as C14orf106, KIAA1903, KNL2, M18BP1) gene (Gene ID 55320) in human. Mis18-binding protein 1 is a component of the Mis18 complex (Mis18α, Mis18β, and M18BP1/KNL2) that plays a role in mediating cell cycle-dependent deposition/incorporation of the histone H3 variant CENP-A at the centromeres. CENP-A incorporation is carried out by at least two sets of events, the Mis18 complex must first localize to centromeres at the onset of anaphase, HJURP-mediated CENP-A deposition then occurs during G1. Polo-like kinase 1 (Plk1) is shown to phosphorylate M18BP1 on Thr702 and plays an important role in promoting Mis18 complex centromere localization.

Specificity

Specifically reacts with the immunogen peptide, but not the corresponding non-phosphorylated sequence, by dot blot analysis. Phosphorylation site is not present in the spliced isoform 2 of human Mis18-binding protein 1/M18BP1.

Anti-phospho Mis18-binding protein 1 (Thr702) Antibody is an antibody against phospho Mis18-binding protein 1 for use in Immunocytochemistry, Dot Blot, Western Blotting.
Immunocytochemistry Analysis: 4.0 µg/mL from a representative lot detected phospho Mis18-binding protein 1 (Thr702) in HeLa cells.
Dot Blot Analysis: 1 µg/mL from a representative lot detected the immunogen peptide, but not the corresponding non-phosphorylated sequence, by dot blot analysis.
Immunocytochemistry Analysis: A representative lot detected Thr702 phosphorylated Mis18-binding protein 1/M18BP1 localized at the centromeres in HeLa cells (McKinley, K.L., and Cheeseman, I.M. (2014). Cell. 158(2):397-411).
Western Blotting Analysis: A representative lot detected in vitro phosphorylation of recombinant Mis18-binding protein 1/M18BP1 on Thr702 by recombinant polo-like kinase 1 (Plk1) (McKinley, K.L., and Cheeseman, I.M. (2014). Cell. 158(2):397-411).

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
