General description

The Mammalian Amino-Terminal FLAG Stable Expression Kit provides the specialized FLAG® components needed for expression of a recombinant N-terminal FLAG fusion in mammalian cells. The pFLAG-CMV-3 and pFLAG-CMV-4 expression vectors encode the aminoglycoside phosphotransferase II gene (Neo) which confers resistance to aminoglycosides such as G418, allowing for selection of stable transfectants.

Application

The pFLAG-CMV-3 and pFLAG-CMV-4 Expression Vectors are suitable for transient or stable expression of secreted or cytoplasmic N-terminal FLAG® fusion proteins.

Components

  • pFLAG-CMV-3a Expression Vector (E8770) 20 μg
  • pFLAG-CMV-4a Expression Vector (E1775) 20 μg
  • pFLAG-CMV-3-BAPa Control Plasmid (C3972) 20 μg
  • pFLAG-CMV-4-BAPa Control Plasmid (C4722) 20 μg
  • C-CMV-24 Reverse Sequencing Primer (P5475) 1 μg
  • N-CMV-30 Forward Sequencing Primer (P5350) 1 μg
  • ANTI-FLAG M2 Monoclonal Antibody (F3165) 200 μg
  • ANTI-FLAG M5 Monoclonal Antibody (F4042) 200 μg
  • ANTI-FLAG M2 Affinity Gel - Freezer Safe (A2220) 1 ml
  • Met-FLAG-BAP Control Protein (P5975) 100 ug
  • FLAG Peptide (F3290) 4 mg

Principle

The FLAG® system is based on the FLAG marker octapeptide (Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys). The FLAG sequence is hydrophilic and the last 5 amino acids (Asp-Asp-Asp-Asp-Lys) represent the target sequence of the protease enterokinase. FLAG fusion proteins are created by cloning the coding sequence of a protein of interest adjacent to the FLAG coding sequence in a FLAG expression vector, followed by expression in the appropriate host cell type.
