General description

Anti-Histone H3 (N-terminal) specifically recognizes human histone H3 (~17kDa). Histone H3/ histone cluster 1 H3 family member a (HIST1H3A) is a core member of the histone family. HIST1H3A is encoded by the gene mapped to human chromosome 6p22.2. The encoded protein is composed of ~136 amino acid.

Immunogen

synthetic peptide corresponding to amino acids 1-20 of human histone H3, conjugated to KLH.

Application

Anti-histone H3 (N-terminal) antibody produced in rabbit has been used in western blotting and native chromatin immunoprecipitation assay.

Biochem/physiol Actions

Histones acts as structural scaffolds for DNA packaging. Histone H3 plays an important role in replication and DNA repair. Histone H3 is heavily post-translationally modified than the other members of histone family. Mutations in the gene leads to cerebellar high-grade gliomas.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8°C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frostfree” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.

Other Notes

In order to obtain the best results using various techniques and preparations, we recommend determining the optimal working dilutions by titration.
